Assuming that you're running a Mac OS 7 - 9.2.2 here: If your second partition will always be mounted at startup, then likely an easy thing for you to do is to create a alias (shortcut) of your game icon and place the icon into the "Startup Items" folder inside of the System Folder.

As long as whatever game you have is always named "game" then it should work OK without needing to replace the icon.

If there are periods where you don't need "game" to launch every time you boot up. You can disable the icon in the Startup Folder using the Control Panel "Extensions Manager". Or drag the alias out of the Startup Folder until you need it again.
